Can the rack be locked onto the ladder?
The bike rack as is does not have any built on locking features. The owner would have to create a homebrew solution to lock the rack in place.

Is it designed so the handlebars wont scratch the trailer?
It depends on the width of the handle bars, they may need to be padded to avoid any damage to the finish of the RV.

My one bike weight 38 lb?
Weight capacity of SKU#85024 is 200 lbs. This ladder rack replaced SKU#65241 from Stromberg that has a weight capacity of 50 lbs. and is listed in our 2015 Master Catalog. The ladder rack from Stromberg clamped on the tubing of the ladder and has no cross member between the arms, hence the low weight threshold. Our ladder bike rack hooks on to the steps and has two cross member attached to both arms.
